If the filter came from an existing tank and the bioload is similar to what the filter was handling before, there should be no issue IMO/IME.FWIW transfering tank water doesn't really help in the grand scheme of bacteria for biological filtration aside from the water being near the correct temperature. The bacteria reside on surfaces and are not free-floating in the water column. This is why surface area is such an important feature of good biological filtration media.
Sand isn't a issue youre good to go
